Your protein sample volume in percent of total column volume (%)
Your protein sample volume to be processed (L)*
Chromatography medium volume (L)
Dynamic binding capacity (g/L)
Quantity to be bound (g)
Safety margin of chromatography medium (%)*
Chromatography medium volume (L)

* in case the resolution is insufficient please increase the amount of chromatography medium

The chromatography medium volume is calculated from the formula:
The chromatography medium volume (L) = Sample volume to be processed (L) / (Sample volume in percent of total column volume (%) / 100)

* refer to general guidelines in the "Column and system for scale" section, see left-hand navigator

The chromatography medium volume is calculated from the formula:
The chromatography medium volume (L) =(Quantity to be bound (g) * (1+ safety margin (%) / 100)) / (Dynamic binding capacity (g/L)

Calculate your packed bed volume, settled chromatography medium volume and slurry volume

Please check that you have sufficient chromatography medium for the required packed bed volume by calculating the appropriate settled chromatography medium volume. Also ensure you have necessary tank capacity for the medium slurry by calculating the slurry volume. When packing Chromaflow columns we recommend to prepare approximately 5% extra slurry to prevent getting air in the column.
Below given compression factors are approximations that can vary slightly with applications.

Column diameter (mm)
Bed height (cm)
Packed bed volume (L)
Compression factor
Settled chromatography medium volume (L)
Slurry concentration (%)
Slurry volume (L)

Compression factor is defined as gravity settled bed volume / packed bed volume

The volumes are calculated from the formulas:

  • Packed bed volume (L) = Column bed height (cm) * crosssectional area (cm2) * x10-3
  • Settled chromatography medium volume (L) = Packed bed volume (L) * compression factor
  • Slurry volume (L) = Settled chromatography medium volume (L) / (slurry concentration (%) * x
    10-2)
