One column purification approach

A convenient one-column purification procedure on SOURCE™ Q is also possible and offers an interesting approach. Crude trityl-on oligonucleotide, from the synthesis (dissolved in 25% ammonia), is loaded directly onto the column. Removal of non-tritylated failure sequences is followed by on-column cleavage of the trityl-group. Purification of the desired product is achieved by applying a linear sodium chloride gradient. The same approach for sample loading (25% ammonia) can also be applied for trityl-off oligonucleotides.
